目的研究乳化异氟醚对大鼠灌胃给药是否产生镇静催眠作用。方法选择健康SD大鼠,采用序贯法测量大鼠灌胃8%的乳化异氟醚后产生催眠作用的半数有效量(median effective dose,ED50)。求得ED50后,选择健康SD大鼠30只,随机分成3组:乳化异氟醚组(Eiso),灌胃8%乳化异氟醚2.15 mL/100 g〔在ED50基础上增加一个公比r(r=0.8序贯法求催眠作用的剂量公比),即ED50/r=2.15 mL/100 g〕;生理盐水组(NS),灌胃生理盐水2.15 mL/100 g;脂肪乳组(IL),灌胃30%脂肪乳2.15 mL/100 g,每组10只。观察大鼠给药后的神经行为学变化,翻正反射发生率(入睡率),睡眠潜伏期、持续期。结果乳化异氟醚灌胃给药对大鼠有镇静催眠的作用;产生催眠作用的ED50为1.72 mL/100 g;2.15 mL/100 g乳化异氟醚(8%)灌胃后,大鼠翻正反射发生率为90%,睡眠潜伏期约为15~20 min,睡眠持续期长达60 min;给药后观察,大鼠能保持良好的自主呼吸,口唇红润,无腹泻等不良反应。结论乳化异氟醚灌胃能对大鼠产生有效的可逆的镇静催眠作用,并且具有良好的安全性。
Objective To investigate whether sedation and hypnosis effect of emulsified isoflurane on intragastric administration in rats. Methods Healthy SD rats were selected and the median effective dose (ED50) of hypnotic effect after 8% emulsified isoflurane administration in rats was measured by sequential method. After obtaining ED50, 30 healthy SD rats were selected and randomly divided into 3 groups: Eiso emulsion, 2.15 mL / 100 g 8% emulsified isoflurane orally (r = 0.8, the dose-equivalent ratio of hypnotic effect for sequential therapy), that is, ED50 / r = 2.15 mL / 100 g]; NS group, saline 2.15 mL / ), Gavage 30% fat emulsion 2.15 mL / 100 g, each group of 10. Results The dosage of emulsified isoflurane administered intragastrically to rats had sedative and hypnotic effects; the ED50 with hypnotic effect was 1.72 mL / 100 g; 2.15 mL / 100 g emulsified isoflurane (8% The incidence of regular reflection was 90%, the latent period of sleep was about 15-20 min, and the duration of sleep was as long as 60 min. After administration, the rats maintained good spontaneous breathing, rosy lips and no adverse reactions such as diarrhea. Conclusion Emulsified isoflurane can effectively and reversibly sedative and hypnotic effect in rats, and has good safety.
