目的:利用近红外漫反射技术和化学计量学的方法对利咽解毒颗粒中黄芩苷含量进行快速定量分析。方法:用积分球漫反射测定样品的近红外漫反射光谱,定量模型的预处理方法为多元散射校正(MSC)和一阶微分处理,波长范围为5 508~5 465cm-1,7 220~7 143cm-1,7 251~7 220cm-1,回归方法为偏最小二乘法(PLS)。结果:定量模型的最佳主因子数为6,内部交叉验证均方差RMSECV为0.046 5,决定系数R2为0.989,外部验证预测均方差RMSEP为0.031,系统精密度RSD为1.2%,方法精密度RSD为2.0%。结论:建立预测模型对利咽解毒颗粒中黄芩苷含量进行测定是可行的,该法快速、简便,结果准确。
OBJECTIVE: To rapidly and quantitatively determine the content of baicalin in Liyan Jiedu Granules by using near-infrared diffuse reflectance spectroscopy and chemometrics methods. Methods: The near-infrared diffuse reflectance spectra of diffuse reflectance spectra were determined by diffuse reflectance of integrating sphere. The pretreatment method of quantitative model was multiscale scatter correction (MSC) and first-order differential processing. The wavelength range was from 5 508 to 5 465 cm -1 and from 1,7 220 to 7 143cm-1,725 ~ 7 220cm-1, the regression method for partial least-squares method (PLS). Results: The best principal factor of the quantitative model was 6, RMSECV of internal cross-validation was 0.046 5, coefficient of determination R2 was 0.989, RMSEP of external validation was 0.031, RSD was 1.2%, and precision RSD Is 2.0%. Conclusion: It is feasible to establish the prediction model to determine baicalin content in Liyan Jiedu Granules. The method is rapid, simple and accurate.
