目的研究高翅果菊Pterocypsela elata根茎中的主要化学成分,探讨lactuside B的抗脑缺血活性。方法采用柱色谱对植物中的提取物进行分离提纯,并利用现代波谱方法分析鉴定,确定化合物的结构;采用大鼠不完全性脑缺血模型,测定大鼠脑组织的含水量、MDA和SOD值,检测lactuside B的抗脑缺血活性。结果从该植物中分离出10个化合物,分别鉴定为莴苣苷B(lactuside B,1)、11β,13-二氢莴苣内酯乙酸酯(2)、β-谷甾醇(3)、胡萝卜苷(4)、(24R)-5α-豆甾-7,22(E)-二烯-3α-醇(5)、3,3′,4-三甲氧基鞣花酸(6)、β-香树脂醇(7)、齐墩果酸(8)、二十六碳酸(9)、硬脂酸(10)。其中化合物1为主要成分,得率0.15%;lactuside B可降低脑组织含水量,降低缺血后脑组织MDA的量,升高SOD的量。结论 10个化合物均为首次从该植物中分离获得,化合物1为主要成分,具有明显的抗脑缺血作用。
Objective To study the main chemical constituents in the roots of Pterocypsela elata and discuss the anti-cerebral ischemic activity of lactuside B. Methods The extracts from plants were isolated and purified by column chromatography. The structure of the compounds was identified by modern spectroscopic analysis. The brain water content, MDA and SOD in rats Value, tested for anti-cerebral ischemic activity of lactuside B. As a result, 10 compounds were isolated from this plant and identified as lactuside B, 1β, 13-dihydro lactamase acetate (2), β-sitosterol (3), daucosterol (4), (24R) -5α-stigmaster-7,22 (E) -dien-3a-ol (5), 3,3 ’, 4-trimethoxylatelane (6) Resin alcohol (7), oleanolic acid (8), hexacosanoic acid (9), stearic acid (10). Among them, compound 1 was the main component with a yield of 0.15%. Lactuside B could decrease the water content of brain tissue, decrease the amount of MDA in brain tissue after ischemia and increase the amount of SOD. Conclusion All the 10 compounds were isolated from this plant for the first time. Compound 1 was the main component and had obvious anti-ischemic effects.
