目的:通过对双莲方抗肝癌细胞SMMC-7721有效组分配伍研究,确定双莲方抗肿瘤有效组分最佳配伍比例。方法:采用CCK-8法检测双莲方中各抗肿瘤有效组分对肝癌细胞SMMC-7721的增殖抑制作用,确定各有效组分对肝癌细胞SMMC-7721的半数抑制浓度(IC_(50)),通过U_(11)(11~6)均匀设计对双莲方抗肿瘤有效组分配伍进行优化,并采用CCK-8法和Hoechst 33358/PI双染色法实验比较和验证优化组分配伍对肝癌细胞SMMC-7721增殖及凋亡的影响。结果:应用均匀设计回归分析得到双莲方最佳配伍比例为半枝莲总黄酮∶半枝莲多糖∶半边莲总黄酮∶半边莲生物碱∶夏枯草总黄酮=4.8∶7.6∶7.8∶13.5∶1。验证实验显示,最佳配伍对肝癌细胞SMMC-7721有较好的增殖抑制和诱导凋亡作用,且其作用优于各组分单用(P<0.05)。结论:通过本实验筛选出的双莲方抗肝癌细胞最佳有效组分配伍抑制肝癌细胞SMMC-7721增殖和诱导其凋亡效果明确,为抗肿瘤新药的研发奠定了基础。
OBJECTIVE: To determine the optimal combination of anti-tumor active ingredients of Shuanglianfang by studying the compatibility of effective components of Shuanglianfang anti-hepatoma SMMC-7721 cells. Methods: The inhibitory effect of various anti-tumor active components of Shuanglianfang on the proliferation of SMMC-7721 cells was detected by CCK-8 assay, and the half inhibitory concentration (IC 50) of each effective component on SMMC-7721 cells was determined. , And the compatibility of anti-tumor effective components of Shuanglianfang was optimized by uniform design of U 11 (11 ~ 6). The CCK-8 method and Hoechst 33358 / PI double staining method were used to compare and verify the compatibility of the optimized components to liver cancer Effect of proliferation and apoptosis of SMMC-7721 cells. Results: The optimal ratio of Shuanglianfang was the total flavonoids of Scutellaria barbata, which was obtained by uniform design regression analysis: polysaccharides of Scutellaria barbata: total flavonoids of Lobelia: Lobelia total flavonoids = 4.8: 7.6: 7.8: 13.5: 1. The validation experiments showed that the best compatibility could inhibit proliferation and induce apoptosis of hepatocellular carcinoma cells SMMC-7721, and the effect was better than that of the other components alone (P <0.05). Conclusion: The optimal combination of anti-hepatoma cells of Shuanglianfang, which was screened by this experiment, can inhibit proliferation and induce apoptosis of SMMC-7721 hepatoma cells, which lays the foundation for the development of new antitumor drugs.
