目的:建立大鼠乳腺癌癌前病变疾病模型,研究阳和化岩汤对大鼠乳腺癌癌前病变的抑制作用。方法:采用DMBA诱导建立大鼠乳腺癌癌前病变模型,9周后分组用药干预治疗4周,观察大鼠体重增长及肿瘤出现情况,取乳腺组织观察病理组织学形态。结果:造模组大鼠肉眼肿瘤发生率92.86%,平均肿瘤个数为4.21,癌前病变和浸润性癌发生率分别21.43%、71.43%,与空白对照组比较具有显著差异(P<0.01)。各治疗组大鼠肉眼肿瘤发生率及平均肿瘤个数与疾病模型组比较,均明显降低和减少(P<0.05);阳和化岩汤组肿瘤发生率为64.71%,平均肿瘤个数为1.82,与其他治疗组比较,无统计学差异(P>0.05)。各治疗组大鼠乳腺组织病理形态学较疾病模型组病变程度轻(P<0.05);阳和化岩汤组癌前病变发生率为47.06%,浸润性癌发生率为29.41%,与其他治疗组比较,无统计学差异(P>0.05)。结论:DMBA小剂量分次给药建立大鼠乳腺癌癌前病变动物模型具有明显优越性,阳和化岩汤为代表的温阳散结中药能够部分抑制乳腺癌癌变,适于乳腺癌高危人群的预防治疗。 Objective: To establish a rat model of precancerous lesions of breast cancer and to study the inhibitory effect of Yanghe Huanyan decoction on precancerous lesions of breast cancer in rats. Methods: The rat model of precancerous lesions of breast cancer was induced by DMBA. After 9 weeks of treatment, the rats were divided into groups and treated for 4 weeks. The growth of the rats and the appearance of tumors were observed. The histopathological changes of the breast tissues were observed. Results: The incidence of macroscopic tumors was 92.86%, the average number of tumors was 4.21, the incidence of precancerous lesions and invasive carcinoma were 21.43% and 71.43% respectively, which was significantly different from that of the blank control group (P <0.01) . The incidence of malignant tumors and the average number of tumors in each treatment group were significantly decreased and decreased compared with the model group (P <0.05). The incidence of tumors in Yanghe Huanyan Decoction group was 64.71%, and the average number of tumors was 1.82 , No significant difference compared with other treatment groups (P> 0.05). The pathological changes of mammary gland tissue in the treatment groups were lighter than that in the disease model group (P <0.05). The incidence of precancerous lesions was 47.06% in Yanghuayan Decoction group and 29.41% in invasive cancer group. Compared with other treatments There was no significant difference between the two groups (P> 0.05). Conclusion: The DMBA small dose and sub-administration of rat model of precancerous lesions of breast cancer has obvious advantages, Yang and Hua Yan Tang, represented by traditional Chinese medicine can partially inhibit the cancer of breast cancer, suitable for high-risk groups of breast cancer Preventive treatment.
