为探讨中国人北方汉族 2型DM醛糖还原酶 (AR)基因 5’端的 (AC)n的多态性与糖尿病视网膜病变易感性的关系 ,用32 PdCTP掺入PCR ,6 %甲酰胺聚丙烯酰胺凝胶电泳和放射性自显影的方法 ,分析了 116例 2型DM〔包括无并发症 (NDC)组 6 6例 ,视网膜病变 (DRP)组 5 0例〕和 42例正常对照者 (CON)AR基因 5′端 (AC)n的多态性。结果显示 ,DM组和对照组均发现 7种等位基因Z - 6Z + 6 ,Z 2等位基因频率在DRP高于NDC和CON组 ;Z+ 2等位基因频率在DRP低于NDC和CON组。结论 :中国北方汉族 2型DM患者AR基因 5’端 (AC)n存在 7种等位基因 (Z - 6Z + 6 )。Z 2等位基因可能是DRP的易感基因 ,Z + 2等位基因可能为其保护基因。
To investigate the association of (AC) n polymorphism at the 5 ’end of type 2 DM aldose reductase (AR) gene with the susceptibility to diabetic retinopathy in Chinese Han population of northern China, PCR was performed using 32 PdCTP incorporation and 6% formamide polypropylene Methods Sixty-six patients with type 2 diabetes (including 66 cases with no-complication (NDC) and 50 cases with retinopathy (DRP)) and 42 normal controls (CON) were analyzed by gel electrophoresis and autoradiography. AR gene 5 ’(AC) n polymorphism. The results showed that the frequencies of allele Z - 6Z + 6 and Z 2 in DR group and DR group were higher than those in NDC and CON group. The allele frequencies of Z + 2 allele in DR group were lower than those in NDC group and CON group . CONCLUSION: There are 7 alleles (Z - 6Z + 6) at the 5 ’end of AR gene in type 2 DM patients in northern China. The Z 2 allele may be a susceptibility gene for DRP, and the Z + 2 allele may be its protective gene.
