目的:建立多烯紫杉醇脂质体药物包封率测定方法。方法:采用HPLC法测定脂质体中多烯紫杉醇的含量及包封率,色谱柱:Diamonsil C18(250mm×4.6 mm,5μm);流动相:乙腈-水(55∶45);柱温:30℃;流速:1.0mL.min-1;紫外检测波长:230nm。结果:多烯紫杉醇在20.4~204μg·mL-1范围内线性关系良好(r=0.9997),平均回收率为97.59%,RSD为1.53%。结论:该方法简单、快速、准确,可用于多烯紫杉醇脂质体包封率的测定。
Objective: To establish a method for the determination of entrapment efficiency of docetaxel liposomes. Methods: The contents and encapsulation efficiency of docetaxel in liposomes were determined by HPLC. Diamonsil C18 (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase was acetonitrile-water (55:45) ℃; flow rate: 1.0mL.min-1; UV detection wavelength: 230nm. RESULTS: Docetaxel had good linearity (r = 0.9997) in the range of 20.4 ~ 204μg · mL-1 with an average recovery of 97.59% and a RSD of 1.53%. Conclusion: The method is simple, rapid and accurate and can be used to determine the entrapment efficiency of docetaxel liposomes.
