吴茱萸水溶性成分HPLC指纹图谱与化学模式识别研究

目的研究吴茱萸药材水溶性化学成分的HPLC指纹图谱,并应用系统聚类分析和主成分分析建立吴茱萸药材的化学模式识别方法,以期为吴茱萸药材质量控制与品质评价提供参考。方法色谱柱为Ultimate XB-C18色谱柱(150.0 mm×4.6 mm,3μm);流动相为乙腈-水(含0.5%醋酸和0.04%磷酸)混合溶液,梯度洗脱;体积流量0.8 mL/min;检测波长为327 nm;进样量10μL;柱温:25℃。结果 34批吴茱萸药材有27个共有峰,相似度在0.945~0.997。聚类分析能对《中国药典》2010年版规定的3种吴茱萸药材进行有效识别,并且从水溶性化学成分上揭示吴茱萸药材的种质远近关系,主成分分析结果与聚类分析结果类似。结论建立的方法能够成功应用于吴茱萸药材的质量评价与控制及品种识别,可为制定吴茱萸质量评价与控制模式提供新的参考依据。 OBJECTIVE To study the HPLC fingerprints of water-soluble chemical constituents from Fructus Evodiae, and to establish a chemical pattern recognition method for Fructus Evodiae by systematic cluster analysis and principal component analysis in order to provide reference for the quality control and quality evaluation of Evodia. The column was Ultimate XB-C18 (150.0 mm × 4.6 mm, 3 μm). The mobile phase consisted of a mixture of acetonitrile and water (containing 0.5% acetic acid and 0.04% phosphoric acid) Detection wavelength of 327 nm; injection volume 10μL; column temperature: 25 ℃. Results 34 batches of Evodia officinalis herbs have 27 common peaks, the similarity of 0.945 ~ 0.997. Cluster analysis can effectively identify the three species of Evodia rutaecarpa stipulated in the 2010 edition of Chinese Pharmacopoeia and reveal the germplasm distance relationship of Evodia rutaecarpa from the water-soluble chemical composition. The principal component analysis results are similar to those of the cluster analysis. Conclusion The established method can be successfully applied to the quality evaluation and control of Evodia rutaecarpa and identification of cultivars, which may provide a new reference for the development of quality control and evaluation of Evodia rutaecarpa.
