目的:通过检测基质金属蛋白酶-7(matrix metalloproteinase-7,MMP-7)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)在胃癌中的表达,探讨它们在肿瘤转移和侵袭过程中的作用。方法:1.收集46例临床病理资料完整的胃癌手术切除标本及10例正常的胃组织标本。2.每例切片3张,分别用于HE染色病理形态学观察和MMP-7和MMP-9的表达情况的检测,分析其与胃癌浸润转移的关系。3.数据统计学分析用SPSS11.0软件,采用χ2检验和Spearman等级相关分析,P<0.05具有统计学意义。结果:10例正常胃组织中MMP-7和MMP-9的表达为阴性,46例胃癌组织中MMP-7的阳性表达率为73.91%,显著高于正常胃组织(P<0.05),MMP-7阳性表达与胃癌淋巴结转移、浸润程度、分化程度相关(P<0.05),与患者性别、年龄无关(P>0.05)。MMP-9的阳性表达率为80.43%,显著高于正常胃组织(P<0.05),MMP-9阳性表达与胃癌淋巴结转移、浸润深度、分化程度相关(P<0.05),与患者性别、年龄无关(P>0.05)。Spearman等级相关分析显示,MMP-7和MMP-9表达呈正相关(相关系数r=0.504,P<0.01)。结论:胃癌组织中MMP-7和MMP-9呈现异常表达。胃癌组织中MMP-7和MMP-9的阳性率与胃癌淋巴结转移、浸润程度、分化程度相关,提示它们在胃癌的发生发展和浸润转移过程中有促进作用。胃癌组织中MMP-7和MMP-9表达正相关,提示其在胃癌的发生发展以及浸润转移中存在着相互促进的作用。MMP-7和MMP-9各自都可以成为胃癌诊断和预后判断的指标。
OBJECTIVE: To detect the expression of matrix metalloproteinase-7 (MMP-7) and matrix metalloproteinase-9 (MMP-9) in gastric carcinoma and to explore their roles in tumor metastasis and invasion Role. Forty-six cases of gastric cancer resected specimens and 10 cases of normal gastric tissues were collected. The results of histological examination and MMP-7 and MMP-9 expression in HE staining were analyzed respectively. Spearman rank correlation analysis showed that MMP-7 and MMP-9 expression was positively correlated (correlation coefficient r = 0.504, P <0.01). Conclusion: The expression of MMP-7 and MMP-9 in gastric cancer is abnormal. The positive rates of MMP-7 and MMP-9 in gastric cancer tissues were correlated with lymph node metastasis, infiltration and differentiation of gastric cancer, suggesting that they may promote the development and invasion of gastric cancer. The positive correlation between the expression of MMP-7 and MMP-9 in gastric cancer suggests that it may promote each other in the occurrence, development and invasion and metastasis of gastric cancer. Each of MMP-7 and MMP-9 can be an indicator of gastric cancer diagnosis and prognosis.
