目的探讨胚胎胸腺组织移植对系统性红斑狼疮(SLE)模型鼠的治疗效果。方法将21只符合要求的BALB/c×C57BL/6F1小鼠经尾静脉注射亲代淋巴细胞混悬液后随机分成3(A~C)组,于SLE模型建立成功后接受相应治疗方案,第25天处死模型小鼠,采用流式细胞术检测各组小鼠外周血中CD4+/CD8+T细胞的比值;电子天平称取小鼠胸腺湿质量后计算胸腺指数;直接免疫荧光法检测肾冰冻切片免疫复合物的荧光强度;间接免疫荧光法检测血清中抗核抗体(ANA)的荧光滴度。结果①C组外周血中CD4+/CD8+T细胞的比值显著高于A组(P<0.05),但A组与B组相比,差异无显著性(P>0.05);②B组胸腺指数与A组相比,差异无显著性(P>0.05),但显著低于C组(P<0.05);③C组肾脏冰冻切片荧光强度及外周血ANA荧光滴度均低于A组和B组(P<0.05)。结论胚胎胸腺移植对SLE模型鼠有显著治疗效果,有望运用于临床SLE的治疗。
Objective To investigate the therapeutic effect of embryonic thymus transplantation on systemic lupus erythematosus (SLE) model rats. Methods Twenty-one eligible BALB / c × C57BL / 6F1 mice were randomly divided into 3 groups (A ~ C) after injection of the parental lymphocyte suspension through tail vein. After successful establishment of SLE model, The mice were sacrificed daily, and the ratio of CD4 + / CD8 + T cells in peripheral blood of each group was detected by flow cytometry. The thymus index was calculated after the electronic balance of thymus was weighed and the frozen sections of kidney were detected by direct immunofluorescence The fluorescence intensity of immunocomplexes was detected by indirect immunofluorescence assay. The fluorescence titer of anti-nuclear antibody (ANA) in serum was detected by indirect immunofluorescence assay. Results ① The ratio of CD4 + / CD8 + T cells in peripheral blood of group C was significantly higher than that of group A (P <0.05), but there was no significant difference between group A and group B (P> 0.05) (P> 0.05), but was significantly lower than that of C group (P <0.05) .③The fluorescence intensity of frozen section and the ANA fluorescence titer of peripheral blood in group C were lower than those in group A and group B <0.05). Conclusion Embryonic thymus transplantation has a significant therapeutic effect on SLE model rats and is expected to be used in the treatment of clinical SLE.
