目的:探讨癌细胞E2F-1因子的表达状态及其与细胞周期的关系。方法:采用免疫荧光组织化学染色法检测8株胃肠道肿瘤细胞系E2F-1转录因子的表达,并采用流式细胞术检测肿瘤细胞周期的变化。结果:依据E2F-1阳性表达的程度不同,将8株肿瘤细胞系分为E2F-1高表达组(包括HGC-27、BGC-823、SGC-7901、HT29)和E2F-1低表达组(包括MKN45、MKN28、SH288、SW-620)。E2F-1高表达组G1/G0期、S期、细胞凋亡比例分别为(33.48±4.61)%、(63.91±5.02)%、(1.50±0.56)%,低表达组分别为(58.58±3.41)%、(35.38±4.03)%、(5.61±1.44)%,2组间比较差异有统计学意义(t分别为6.04、5.99和2.44,P<0.05或P<0.01)。结论:肿瘤细胞E2F-1因子的表达状态对细胞周期有明显的影响。E2F-1阳性的肿瘤细胞则倾向于更高的增殖速率。
Objective: To investigate the expression of E2F-1 in cancer cells and its relationship with cell cycle. Methods: The expression of E2F-1 transcription factor in 8 gastrointestinal cancer cell lines was detected by immunofluorescence staining, and the changes of cell cycle were detected by flow cytometry. Results: According to the degree of E2F-1 positive expression, 8 tumor cell lines were divided into E2F-1 high expression group (including HGC-27, BGC-823, SGC-7901, HT29) and E2F-1 low expression group ( Including MKN45, MKN28, SH288, SW-620). The percentages of G1/G0 phase, S phase and apoptosis in E2F-1 high expression group were (33.48±4.61)%, (63.91±5.02)%, (1.50±0.56)%, respectively, and those in low expression group were (58.58±3.41). )%, (35.38±4.03)%, (5.61±1.44)%, the difference between the two groups was statistically significant (t = 6.04, 5.99, and 2.44, respectively, P <0.05 or P <0.01). Conclusion: The expression of E2F-1 in tumor cells has a significant effect on cell cycle. E2F-1 positive tumor cells tend to have higher proliferation rates.
