传统的免疫共沉淀测序(ChIP-seq)是研究蛋白-染色质相互作用的主流方法,由于ChIP-seq难以在低起始细胞量和单细胞样本上实施限制了其应用范围。近年来新发展起来的染色质酶切测序技术CUT&RUN和CUT&Tag展现出操作简单,背景低,可重复性高和要求的细胞起始量低等优点,有替代ChIP-seq进行蛋白-染色质相互作用研究的潜力,并有效提高了单细胞ChIP-seq的可行性。本文将就这类靶向染色质进行酶切测序的主要技术的发展和应用现状做简要综述。“,”Chromatin immunoprecipitation followed by sequencing (ChIP-seq) is the most popular method for the study of protein-chromatin interaction . However, it is difficult to implement on a low start-ing number of cells or single-cell application. The chromatin cleavage under target sequencing technology emerged in recent years has shown advantages such as simple operation, low background, high reproducibi-lity, and low starting cell numbers. It can replace the traditional ChIP-seq and improve single-cell Chip-seq method. This article briefly reviews the development and application situation of this type of chromatin cleavage under target sequencing technology.
