目的:分析并比较卵巢癌细胞中与肿瘤转移、侵袭相关的转录因子。方法:采用细胞划痕及小室跨膜实验比较卵巢癌细胞HO-8910及其高转移株HO-8910PM的转移侵袭能力差异;采用高通量转录因子活性检测芯片(DNA/蛋白结合芯片)分析并比较两种细胞中转录因子的活化情况,明确与肿瘤侵袭、转移相关表达的转录因子;用转录因子酶联免疫活性分析方法验证芯片检测结果。结果:细胞划痕实验及侵袭实验表明HO-8910PM比HO-8910细胞转移侵袭能力强,在345个转录因子中共筛选出与肿瘤侵袭、转移相关的31个表达差异转录因子。HO-8910PM细胞与HO-8910细胞相比,表达上调的有13种,下调的有18种。v-Maf仅表达于HO-8910PM,HLF仅表达于HO-8910。结论:v-Maf、HLF等31种转录因子可能参与了卵巢癌侵袭转移的相关机制。
Objective: To analyze and compare the transcription factors related to tumor metastasis and invasion in ovarian cancer cells. METHODS: The cell invasion and invasion of ovarian cancer cell line HO-8910 and its highly metastatic HO-8910PM cells were compared by cell scratch assay and transmembrane transmembrane assay. High-throughput transcription factor activity assay (DNA / protein-binding chip) The activation of transcription factors in both cells was compared, and the transcription factors related to tumor invasion and metastasis were determined. The results of chip detection were verified by the transcription factor enzyme-linked immunosorbent assay. Results: The results of cell scratch assay and invasion assay showed that HO-8910PM had stronger metastatic potential than HO-8910 cells, and 31 out of 345 transcription factors were screened for 31 differentially expressed transcription factors related to tumor invasion and metastasis. HO-8910PM cells compared with HO-8910 cells, 13 were up-regulated, 18 down-regulated. v-Maf is expressed only on HO-8910PM and HLF is only expressed on HO-8910. CONCLUSION: Thirty-one transcription factors such as v-Maf and HLF may be involved in the mechanism of invasion and metastasis of ovarian cancer.
