目的 为进一步探讨细胞机构感受机制 ,建立一套实用的培养细胞牵张刺激模拟装置。方法 用有机玻璃和可变形膜设计制作一套培养细胞牵张刺激模拟装置 ,并用同位素标记氨基酸和核苷酸掺入法作初步应用评价。结果 应用自行设计制作的两个模型 ,对培养细胞进行较强的牵张刺激 ,发现 2 0 %的拉长或 12kPa负压的球面牵张对培养心肌细胞的数量和乳酸脱轻酶释放均无影响 ;但可显著刺激培养心肌细胞3H 亮氨酸及成纤维细胞3H TdR的掺入。结论 初步应用说明该装置结构简单实用
Objective To further explore the mechanism of cellular mechanism and to establish a practical model of cell stimulation stimulator. Methods A set of simulating devices for cell stretch stimulation of cultured cells was designed and manufactured with plexiglass and deformable membrane. The preliminary application of isotope labeled amino acids and nucleotide incorporation was evaluated. Results Two models designed by ourselves were used to stimulate the cultured cells. The results showed that the number of cultured cardiomyocytes and the release of lactate-removing-light enzyme were all decreased with 20% stretch or 12-kPa negative pressure spherical stretch But could significantly stimulate the incorporation of 3H-leucine in cardiac myocytes and 3H-TdR in fibroblasts. Conclusion Preliminary application shows that the device is simple and practical
