目的 :探讨 MDM2基因在鼻咽癌 (NPC)中的表达情况及其与 p5 3蛋白表达、EB病毒 (EBV )潜伏感染的关系。方法 :运用非同位素原位杂交、免疫组化和多聚酶链反应技术对 46例 NPC、12例慢性鼻咽炎症粘膜(CINE)分别进行 MDM2 m RNA、MDM2蛋白、p5 3蛋白及 EBV- DNA的检测。结果 :46例 NPC中 ,14例为MDM2 m RNA及 MDM2蛋白高表达 ,38例为 p5 3蛋白阳性 ,43例为 EBV- DNA阳性 ;12例 CINE均无 MDM2m RNA、MDM2蛋白及 p5 3蛋白表达 ;MDM2的高表达与 p5 3蛋白的表达相关 (P <0 .0 5 ) ,而与 EBV- DNA无关。结论 :MDM2可能通过与 p5 3蛋白相互作用而在 NPC的发生过程中起着重要作用。
Objective: To investigate the expression of MDM2 gene in nasopharyngeal carcinoma (NPC) and its relationship with the expression of p5 protein and latent infection with Epstein-Barr virus (EBV). Methods: MDM2 m RNA, MDM2 protein, p5 3 protein and EBV-DNA in 46 cases of NPC and 12 cases of chronic nasopharyngeal inflammatory mucosa (CINE) were detected by using non-isotope in situ hybridization, immunohistochemistry and polymerase chain reaction . Results: Of the 46 NPC cases, MDM2 m RNA and MDM2 protein were highly expressed in 14 cases, 38 cases were positive for p5 3 protein and 43 cases were EBV-DNA positive. No MDM2 mRNA, MDM2 protein and p5 3 protein were found in 12 cases of CINE ; MDM2 high expression and p5 3 protein expression (P <0 05), and has nothing to do with EBV-DNA. Conclusion: MDM2 may play an important role in the pathogenesis of NPC through the interaction with p5 3 protein.
