研究白细胞介素 10(rhIL-10)对白细胞介素1β(rhIL-1β)诱导的人肾小球系膜细胞(human mesangialcells,hMsC)的细胞间粘附分子1(CD54)表达的影响及其细胞内作用机制。方法:用细胞ELISA法检测CD54蛋白水平表达,以 Northern杂交方法测定 CD54的基因表达,采用电泳迁移率变动法观察核因子κB(nuclear factor κB,NFκb)的活性。结果:20ng·L~(-1)的rhIL-10能抑制rhIL-1β诱导的CD54的表达,此抑制作用在rhIL-1β加入后的36h内持续存在,rhIL-10还能抑制 rhIL-1β诱导的 hMsC CD54基因表达和核因子κB活化。结论:细胞因子 rhIL-10对rhIL-1β诱导的hMsC的CD54表达的抑制作用至少部分与其对NFκB活化及CD54基因表达的抑制有关。
To investigate the effect of interleukin-10 on the expression of intercellular adhesion molecule-1 (CD54) in human mesangial cells induced by rhIL-1β Intracellular mechanism. Methods: The expression of CD54 was detected by ELISA. The expression of CD54 was detected by Northern blot. The activity of nuclear factor κB (NFκB) was detected by electrophoretic mobility shift assay. Results: 20ng · L -1 of rhIL-10 could inhibit the expression of CD54 induced by rhIL-1β, which could be observed within 36h after the addition of rhIL-1β. RhIL-10 also inhibited the induction of rhIL-1β HMsC CD54 gene expression and nuclear factor kappa B activation. CONCLUSION: The inhibitory effect of rhIL-10 on rhIL-1β-induced CD54 expression in hMsC is at least partly related to the inhibition of NFκB activation and CD54 gene expression.
