目的寻求一种简单易行的观察视网膜神经节细胞形态学的染色方法。方法取30d(P30)Long Even’s大鼠6只,雌雄不限,麻醉处死,取眼球后剥离完整视网膜,置于4%多聚甲醛溶液中固定12h,取固定好的视网膜行全视网膜铺片后DiI荧光染色,将视网膜再次置于4%多聚甲醛溶液中避光37℃固定,6d后观察神经节细胞形态。结果染色可见神经节细胞胞体清晰,树突及轴突均匀着色,形态清晰。结论视网膜铺片行DiI染色可以较好的观察神经节细胞形态。
Objective To find a simple and easy method for observing the morphology of retinal ganglion cells. Methods Six Long Even’s rats (30 days old) were randomly divided into male and female rats. After anesthesia was sacrificed, the intact retina was peeled off and placed in 4% paraformaldehyde solution for 12 hours. After the retina was fixed, DiI fluorescence staining, the retina was placed again in 4% paraformaldehyde solution 37 ° C fixed light, 6d after the observation of ganglion cell morphology. Results showed that the ganglion cell bodies were clearly stained, the dendrites and axons were evenly colored and the morphology was clear. Conclusion DiI staining of retinal sheets can better observe the morphology of ganglion cells.
