目的 观察不同剂量、辐照时间高频诊断超声(HFDU)对兔晶状体上皮细胞(RLEC)超微结构和细胞周期的影响。 方法 采用频率15MHz、热力指数(Ti)为0、机械指数(Mi)为0 .1和0 .37的HFDU经眼直接法,辐照4 8只新西兰兔左眼晶状体15min和30min ,分别于辐照后12 ,2 4和4 8h取其前囊膜作电镜观察RLEC的超微结构,流式细胞术检查RLEC的细胞周期;取未辐照的右眼RLEC为对照。 结果 (1)HFDU对RLEC的超微结构有一定影响,随着辐照时间延长或Mi增加结构损伤加重。Mi0 .1辐照15min后12h ,多数RLEC结构正常;辐照15min或30min后2 4h ,主要表现为线粒体和粗面内质网变化。Mi 0 .37辐照30min后,RLEC出现早期凋亡现象。对照眼未见上述变化。(2 )HFDU引起RLEC细胞周期各期比例再分布,随着辐照时间延长或Mi增加可出现凋亡峰。 结论 HFDU可引起RLEC超微结构变化、细胞周期各期比例的再分布及细胞凋亡,这种改变随辐照时间延长或Mi的增高而加重。眼科超声检查时使用低Mi(0 .1)、检查时间<15min较为安全。
Objective To observe the effect of HFDU on the ultrastructure and cell cycle of rabbit lens epithelial cells (RLECs) with different doses and irradiation time. Methods Forty-eight New Zealand rabbits were irradiated with HFDU by ocular direct method for 15min and 30min at 15MHz with Ti (Ti) of 0 and mechanical index (Mi) of 0.1 and 0.37. The ultrastructure of RLECs was observed by electron microscopy at 12, 24, and 48 hours after transplantation. The cell cycle of RLECs was examined by flow cytometry. The RLEC of non-irradiated right eyes was used as control. Results (1) The HFDU had some influence on the ultrastructure of RLEC. With the extension of irradiation time or the increase of structural damage, Mi increased. Mi0.1 irradiation 15min after 12h, most of the RLEC normal structure; irradiation 15min or 30min after 24h, mainly for mitochondria and rough endoplasmic reticulum. Mi 0.37 irradiation 30min, RLEC early apoptosis phenomenon. The control eye did not see the above changes. (2) HFDU caused redistribution of the proportion of RLEC cell cycle phases, with the increase of irradiation time or Mi may appear apoptosis peak. Conclusion HFDU can induce ultrastructural changes of RLEC, the redistribution of cell cycle and cell apoptosis, which is aggravated with the prolongation of irradiation time or the increase of Mi. Ophthalmological ultrasonography using low Mi (0.1), check the time <15min is more secure.
