目的检测调节性T细胞中Foxp3在原因不明的复发性流产(URSA)患者的外周血与蜕膜中的表达,探讨Foxp3在免疫耐受中的作用及与复发性流产的关系。方法选择2012年3月—2013年3月就诊的原因不明复发性流产患者28例为URSA组,正常早孕要求终止妊娠的妇女30人为正常组。收集两组患者外周血液及蜕膜组织标本。采用半定量、实时荧光定量PCR分析外周血及蜕膜中Foxp3 mRNA相对含量。结果 Foxp3转录水平在复发性流产患者的外周血及蜕膜组织中明显低于正常对照组(P<0.01)。结论 Foxp3在维持早期妊娠中可能有一定作用,其表达下调可能与复发性流产的发生有关。
Objective To detect the expression of Foxp3 in peripheral blood and decidua of patients with recurrent spontaneous abortion (URSA) of unknown cause in regulatory T cells and to explore the role of Foxp3 in immune tolerance and its relationship with recurrent spontaneous abortion. Methods 28 patients with unexplained recurrent spontaneous abortion (URSA) were selected from March 2012 to March 2013. 30 women with normal pregnancy expectant termination of pregnancy were normal group. Two groups of patients with peripheral blood and decidual tissue samples were collected. Semi-quantitative real-time PCR was used to analyze the relative content of Foxp3 mRNA in peripheral blood and decidua. Results Foxp3 transcription level in patients with recurrent spontaneous abortion in the peripheral blood and decidual tissue was significantly lower than the normal control group (P <0.01). Conclusions Foxp3 may play a role in maintaining early pregnancy. The down-regulation of Foxp3 may be related to the occurrence of recurrent spontaneous abortion.
