目的评价出生后早期低剂量狄氏剂(Dieldrin)暴露对青春期和成年期ICR小鼠黑质纹状体胶质纤维酸性蛋白(GFAP)表达水平的影响。方法仔鼠自出生后(PND)随机分成5个剂量组,即狄氏剂处理组(20、2、0.2μg/kg)、溶剂对照组(DMSO)和生理盐水对照组(NS),于出生后第3天进行腹腔注射染毒,隔天1次,连续6次。分别于青春期和成年期处死一批动物,用Western blot法检测黑质纹状体GFAP的表达情况。结果小鼠出生后早期接触低剂量狄氏剂可引起黑质纹状体GFAP蛋白水平呈剂量依赖性增高,除了0.2μg/kg组和2μg/kg青春期雌鼠组外,其它剂量组均显著升高GFAP蛋白水平,且成年期明显高于青春期。结论出生后早期接触低剂量狄氏剂可引起黑质纹状体GFAP进行性升高。
Objective To evaluate the effect of early low birth Dieldrin exposure on the expression of glial fibrillary acidic protein (GFAP) in substantia nigrostriatal (ICR) mice during adolescence and adulthood. Methods The pups were randomly divided into five groups after birth (PND): dieldrin (20, 2, 0.2μg / kg), solvent control group (NS) and normal saline control group After the first 3 days of intraperitoneal injection of poisoning, every other day 1, 6 consecutive times. Animals were sacrificed at puberty and adulthood respectively. The expression of GFAP in nigrostriatum was detected by Western blot. Results In the early postnatal period, mice exposed to low dose of dieldrin caused a dose-dependent increase in the level of GFAP protein in the nigrostriatal tissue. Except for the 0.2μg / kg group and the 2μg / kg group of adolescent females, the other dose groups significantly increased High GFAP protein levels, and adulthood was significantly higher than puberty. Conclusions The early postnatal exposure to low-dose dieldrin can cause progressive elevation of nigrostriatal GFAP.
