阳江高本底地区居民8-羟基脱氧鸟苷及其修复基因8-羟基鸟嘌呤DNA糖苷酶表达水平

目的探讨小剂量长期连续天然放射性照射人群的8-羟基脱氧鸟苷(8-OHdG)及其修复基因8-羟基鸟嘌呤DNA糖苷酶(hOGG1)表达水平。方法选择我国阳江天然放射性高本底辐射地区(HBRA)50名50~59岁男性居民为研究对象,另选择在恩平市某镇(CA)出生并居住,年龄相仿的50名男性居民为对照人群。采集周围血10 ml,分离血浆和白细胞,酶联免疫吸附实验(ELISA)测定血浆中DNA氧化损伤指标8-OHdG的水平,实时定量基因扩增荧光检测系统测定白细胞中DNA氧化损伤修复基因hOGG1 mRNA相对表达水平。结果与CA组比较,HBRA组人群周围血血浆中DNA氧化损伤指标8-OHdG水平较低,白细胞中hOGG1基因mRNA的相对表达水平较高,以上指标在2组间的差异均有统计学意义(P<0.05)。结论小剂量长期连续照射可能会提高机体DNA氧化损伤修复能力。 Objective To investigate the expression of 8-hydroxydeoxyguanosine (8-OHdG) and its repair gene 8-oxoguanine DNA glycosylase (hOGG1) in low-dose long-term continuous natural radioactive irradiation. Methods 50 male residents aged 50-59 years from HBR in Yangjiang of China were selected as study subjects. Fifty male residents of the same age who were born and lived in a town of Enping City (CA) were selected as the control group . The peripheral blood was collected 10 ml, plasma and leukocyte were separated, the level of 8-OHdG in plasma was detected by enzyme-linked immunosorbent assay (ELISA), and the hOGG1 mRNA was detected by real-time quantitative gene amplification fluorescence detection system Relative expression level. Results Compared with CA group, the level of 8-OHdG, a marker of DNA oxidative damage, in peripheral blood of HBRA group was lower than that in CA group, and the relative expression level of hOGG1 mRNA in leukocytes was higher than that of CA group P <0.05). Conclusion Small doses of long-term continuous irradiation may improve the ability of DNA oxidative damage repair.
