目的:以银杏叶为原料,研究纤维素酶-乙醇协同提取总黄酮的浸提条件,并对粗提物的自由基清除活性进行测定。方法:通过单因素实验和响应面分析法确定最佳提取工艺参数。结果:纤维素酶用量100 U/mL,醋酸-醋酸钠缓冲液pH 4.8,酶解温度50℃,酶解时间2 h,该工艺参数下总黄酮得率为1.53%。1,1-二苯基-2-三硝基苯肼(DPPH.)自由基清除实验表明,在总黄酮浓度为0.05 mg/mL,37℃保温15 min条件下,对0.04 mg/mLDPPH.自由基的清除率可达85.7%。结论:纤维素酶不破坏银杏叶总黄酮有效成分,可以用于银杏叶总黄酮的提取。
OBJECTIVE: To study the extraction condition of flavonoids extracted by cellulase-ethanol using Ginkgo biloba as raw material, and to determine the free radical scavenging activity of the crude extracts. Methods: The optimum extraction parameters were determined by single factor experiment and response surface analysis. Results: The yield of total flavonoids was 1.53% under the conditions of 100 U / mL of cellulase, pH 4.8 of acetic acid-sodium acetate buffer, 50 ℃ of enzymolysis temperature and 2 h of enzymolysis time. 1,1-diphenyl-2-trinitrophenylhydrazine (DPPH.) Free radical scavenging experiments showed that the total flavonoid concentration of 0.05 mg / mL, 37 ℃ incubated 15 min conditions, 0.04 mg / mLDPPH. Free Base clearance rate of up to 85.7%. Conclusion: Cellulase does not destroy the active ingredients of total flavonoids of Ginkgo biloba leaves, which can be used for the extraction of total flavonoids from Ginkgo biloba leaves.
