硕大利什曼原虫感染的小鼠是现在公认的研究Th细胞分化的模型。分别用前鞭毛体以后足垫注射法感染易感型BALB/c小鼠和耐受型C57BL/6小鼠。每组动物于感染同时分别注射抗CD80单抗和抗CD86单抗,之后每隔一天注射抗B7抗体,共4周,处死小鼠后取腹股沟和腘窝淋巴结制备单细胞悬液,加入前鞭毛体共同培育后取上清定量分析IL-4、IL-10和IFN-γ浓度。取被感染的足垫匀浆作有限稀释,计数原虫数。
Leishmania infection in mice is now a well-established model for studying Th cell differentiation. Susceptible BALB / c mice and tolerant C57BL / 6 mice were infected with promastigotes by footpad injection. Each group of animals were injected with anti-CD80 monoclonal antibody and anti-CD86 monoclonal antibody respectively, and then injected with anti-B7 antibody every other day for 4 weeks. After the mice were sacrificed, single-cell suspension was prepared from inguinal and popliteal lymph nodes, After co-cultivation, supernatants were collected for quantitative analysis of IL-4, IL-10 and IFN-γ concentrations. Take infected foot pad homogenate for limited dilution, count the number of protozoa.
