目的 :观察多粘菌素B(PMX B)对大鼠内毒素血症血浆中内毒素 (LPS)和肿瘤坏死因子 α(TNF α)水平的影响。方法 :实验分为 :LPS +PMX B组 ;LPS +生理盐水 (NS)组 ;LPS模型组。给大鼠一次LPS 0 5mg·kg-1,iv ,前 2组分别于注入LPS后 0 5h给予PMX B 0 2mg·kg-1和 1mLNS ,iv ,LPS模型组大鼠于注入LPS后不作其他处理。在给LPS后 0 5 ,1,2 ,4h抽血 ,用鲎试剂检测血浆中LPS ,ELISA法测定TNF α水平。结果 :LPS +PMX B组 ,血浆LPS ,TNF α水平明显下降 ,平均动脉压 (MAP)于 2 ,4h明显高于LPS +NS组和LPS模型组。结论 :PML B治疗能明显降低血浆中LPS ,TNF α水平。
Objective: To observe the effect of polymyxin B (PMX B) on plasma endotoxin (LPS) and tumor necrosis factor α (TNF α) in rats with endotoxemia. Methods: The experiment was divided into: LPS + PMX B group; LPS + saline group; LPS model group. Rats were given LPS 0 5mg · kg-1, iv, the first 2 groups were given PMX B 0 2mg · kg-1 and 1mLNS 0 h after injection of LPS, iv, LPS model rats were injected with LPS after no other treatment . Blood was drawn at 0, 1, 2, and 4 hours after LPS administration, and LPS was used to detect plasma LPS. ELISA was used to determine the level of TNFα. Results: The levels of LPS and TNFα in LPS + PMX B group were significantly decreased. Mean arterial pressure (MAP) at 2 and 4 hours was significantly higher than that in LPS + NS group and LPS model group. Conclusion: PML B treatment can significantly reduce plasma LPS, TNF α levels.
