目的:观察肥胖症相关(fat mass and obesity associated,FTO)基因的rs9939609单核苷酸多态性,并探讨其与多囊卵巢综合征间的关联及与肥胖间的交互作用。方法:采用多聚酶链反应-限制性片段长度多态性方法,分析142例多囊卵巢综合征患者和136名正常对照者的FTO基因型。采用非条件Logistic分析各基因型与多囊卵巢综合征发病易感性间的关系及其与肥胖间的交互作用。结果:携带C(CC/CT)基因个体的患病风险较非C基因携带者(TT)明显增加(OR=1.21,95%CI为1.08~2.61,P<0.01;校正OR=1.37,95%CI为1.27~2.91,P<0.01)。非条件Logistic分析表明,携带CT/CC基因型肥胖个体的多囊卵巢综合征罹患风险是携带TT基因非肥胖个体的3.33倍(OR=3.33,95%CI为2.34~5.01,P<0.01)(相对超额危险度比=1.66,95%CI:1.27~2.19;归因比=0.49,95%CI为0.36~0.79;交互作用指数=1.27,95%CI为1.11~1.94)。结论:FTO rs9939609单核苷酸多态性与多囊卵巢综合征罹患风险增加有关,且与肥胖者的多囊卵巢综合征发病间存在协同效应。
OBJECTIVE: To observe rs9939609 single nucleotide polymorphism of the fat mass and obesity associated (FTO) gene and to explore its association with polycystic ovary syndrome and its interaction with obesity. METHODS: FTO genotypes were analyzed in 142 women with PCOS and 136 healthy controls by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Non-conditional logistic analysis of each genotype and the incidence of polycystic ovary syndrome susceptibility and its interaction with obesity. Results: The prevalence of C (CC / CT) individuals was significantly higher than that of non-C carriers (OR = 1.21, 95% CI 1.08-2.61, P <0.01; OR = 1.37, 95% CI was 1.27-2.91, P <0.01). Non-conditional logistic analysis showed that the risk of PCOS with obese individuals with CT / CC genotypes was 3.33 fold (OR = 3.33, 95% CI, 2.34-5.01, P <0.01) for individuals with TT genes The relative excess risk ratio = 1.66, 95% CI: 1.27 to 2.19; the attributable ratio = 0.49, 95% CI 0.36 to 0.79; interaction index = 1.27, 95% CI 1.11 to 1.94). CONCLUSIONS: FTO rs9939609 SNP is associated with an increased risk of PCOS and a synergistic effect with the incidence of polycystic ovary syndrome in obese subjects.
