目的观察葡萄糖-6-磷酸酶多克隆抗体(G-6-PpAb)对大鼠血管源性脑水肿(VBE)的治疗作用。方法将60只Wistar大鼠随机分为正常、脑水肿、脑水肿甘露醇、脑水肿G-6-PpAb共4组。Wistar大鼠腹腔注射苯肾上腺素造成VBE模型,然后分别股静脉注射甘露醇、腹腔注射G-6-PpAb,用Evansblue(EB)法测定血脑屏障(BBB)的通透性,并以远红外水分分析仪分别测定各组脑灰、白质水分含量百分比。结果G-6-PpAb组同甘露醇组相比对降低BBB通透性及大脑白质水分含量均有显著效果(P<0.01),对灰质的脱水作用无显著性差异(P>0.05)。结论G-6-P活性与VBE中BBB的通透性改变有关,G-6-PpAb对VBE尤其是脑白质水肿有选择性治疗作用。
Objective To observe the therapeutic effect of glucose-6-phosphatase polyclonal antibody (G-6-PpAb) on vasogenic brain edema (VBE) in rats. Methods Sixty Wistar rats were randomly divided into 4 groups: normal, brain edema, brain edema mannitol and brain edema G-6-PpAb. Phenylephrine was injected intraperitoneally into Wistar rats to induce VBE model. Then, mannitol was injected into femoral vein and G-6-PpAb was injected intraperitoneally. The permeability of BBB was measured by Evansblue (EB) Moisture analyzer were measured in each group brain ash, white matter moisture content percentage. Results Compared with the mannitol group, the G-6-PpAb group had a significant effect on reducing BBB permeability and white matter moisture (P <0.01), and had no significant difference on the dehydration of gray matter (P> 0.05). Conclusion G-6-P activity is related to the change of BBB permeability in VBE. G-6-PpAb has a selective therapeutic effect on VBE, especially white matter edema.
