目的 :观察自由基对培养的人胚软骨细胞损伤及硒的防护作用。方法 :在体外培养的人胚关节软骨细胞培养液中 ,加入黄嘌呤、黄嘌呤氧化酶及微量元素硒。测定软骨细胞DNA和蛋白质及软骨基质中蛋白多糖和胶原的含量。结果 :培养 14 4h后 ,软骨细胞DNA、蛋白质及软骨基质中蛋白多糖含量都明显减少 ,但胶原含量无明显改变 ,含硒组减少程度轻。结论 :自由基可能和退形性关节疾病病因有关 ,是否选择微量元素硒预防及治疗自由基引起的软骨损伤需动物实验的进一步研究。
Objective: To observe the protective effects of free radicals on cultured human chondrocytes and selenium. Methods: Xanthine, xanthine oxidase and trace element selenium were added to cultured human embryonic articular chondrocytes in vitro. Determination of chondrocyte DNA and protein and cartilage matrix proteoglycan and collagen content. Results: The content of proteoglycan in DNA, protein and cartilage matrix of chondrocytes decreased significantly after 14 4 h incubation, but the content of collagen did not change significantly, and the reduction in selenium group was slight. CONCLUSION: Free radicals may be related to the etiology of degenerative joint disease. Whether the use of trace element selenium can prevent or treat free radical induced cartilage injury needs to be further studied in animal experiments.
