对结肠定位释药系统(CDDS)的分类,理论依据,辅料应用,制备方法及体内外评价方法等相关研究进展作一综述。在查阅大量国内外文献基础上,进行归纳总结,得出新近研究进展。结肠定位释药系统主要分为酶降解型、pH依赖型、时间依赖型、压力控制型释药系统及2种释药机制联合应用型等;多糖等辅料现多联合使用或对其进行结构修饰以提高结肠定位释药性;除片剂外,微粒、微丸、微球、微囊及纳米制剂的研制成为热点。以结肠部位的生理特征为基础的结肠定位释药系统在结肠及全身疾病的治疗中发挥巨大优势,受到广泛关注。
This review summarizes the classification, theoretical basis, application of excipients, preparation methods and in vitro and in vivo evaluation methods of colon-specific drug delivery system (CDDS). On the basis of consulting a large number of domestic and foreign literatures, this paper summarizes and concludes the recent research progress. Colon-specific drug delivery system is divided into enzymatic degradation, pH-dependent, time-dependent, pressure-controlled drug delivery system and the combination of two kinds of drug release mechanism; polysaccharide and other excipients are now more joint use or its structural modification In order to improve the colonic release drug release; in addition to tablets, the development of particles, pellets, microspheres, microcapsules and nano-formulations become hot spots. The colon-based drug delivery system based on the physiological characteristics of the colon region has gained great advantages in the treatment of colon and systemic diseases and has drawn wide attention.
