采用反相高效液相色谱法(RP-HPLC)对广东水牛乳乳清蛋白成分进行了分离和定量分析研究。结果表明乳清蛋白得到较好的分离且重复性良好,3种主要蛋白质(α-La、β-Lg、BSA)回归方程的线性关系>99%,且其保留时间和峰面积的RSD分别小于0.443和2.67,加标回收率范围为99.5%~101%。在相同条件下对南方水牛乳及荷斯坦牛乳进行差异性分析,表明两种乳源乳清蛋白在组分含量和遗传变异体方面均有明显的差异。
The components of buffalo whey protein in Guangdong buffalo were separated and quantitatively analyzed by RP-HPLC. The results showed that the whey protein was well separated and reproducible. The linear relationship of the regression equation of the three major proteins (α-La, β-Lg, BSA) was> 99%, and the RSDs of retention time and peak area were less than 0.443 and 2.67, the spiked recoveries ranged from 99.5% to 101%. Under the same conditions, the differences of southern milk water and Holstein milk were analyzed, which showed that there were significant differences between the two milk whey proteins in the content of components and the genetic variants.
