目的∶研究中药白花前胡的提取物PPD对野百合碱(MCT)所致大鼠肺动脉高压的影响。方法∶用MCT制造大鼠肺动脉高压模型;以硝苯地平(Nif)为阳性对照药;PV-1聚乙烯管插管测量肺动脉压力;肺组织切片HE染色显微测量肺小动脉中膜厚度;免疫荧光法观察肺组织中Tenascin-C合成的变化。结果∶PPD可明显减少MCT引起的肺动脉压升高,而对体循环压力无明显影响;可明显减少MCT所致的右心指数的升高;可明显减轻MCT所致的肺血管损伤与肺血管重建(明显减少了炎性细胞浸润、TN-C的表达及血管平滑肌细胞的增殖)。结论∶PPD可明显抑制MCT所致大鼠肺动脉高压的形成。
OBJECTIVE: To study the effect of PPD, an extract of Radix Paeoniae Alba, on pulmonary arterial hypertension induced by monocrotaline (MCT) in rats. METHODS: Rat pulmonary hypertension model was established with MCT; nifedipine (Nif) was used as positive control; pulmonary artery pressure was measured with PV-1 polyethylene tube intubation; lung arterial medial thickness was measured with HE staining. The changes of Tenascin-C synthesis in lung tissue were observed by immunofluorescence. RESULTS: PPD could significantly reduce the increase of pulmonary arterial pressure caused by MCT, but had no significant effect on systemic circulation pressure; it could significantly reduce the increase of right heart index caused by MCT; it could significantly reduce the pulmonary vascular injury and pulmonary vascular remodeling caused by MCT. (Significantly reduced inflammatory cell infiltration, TN-C expression, and proliferation of vascular smooth muscle cells). Conclusion: PPD can significantly inhibit the formation of pulmonary hypertension induced by MCT in rats.
