单核细胞趋化蛋白诱导蛋白-1(MCPIP1)是最近发现的一类具有免疫调节作用的CCCH型锌指家族分子。MCPIP1可被LPS、IL-1β或MCP-1等多种炎性因子刺激表达,可通过下调炎症因子(如IL-6、IL-12p40等)表达,从而负向调控炎症过程。MCPIP1的作用机制主要是作为RNA酶调节某些炎性因子mRNA或pre-miRNA的降解。此外,MCPIP1也可作为去泛素化酶靶向TNF受体相关蛋白(TRAFs)成员,从而负向调控JNK和NF-κB信号活化。将从MCPIP1分子的发现、基因和蛋白质结构、生物学功能、表达调控以及临床应用前景等几个方面进行阐述。
Monocyte Chemotactic Protein Inducer Protein-1 (MCPIP1) is a recently discovered class of CCCH-type zinc finger family molecules with immunomodulatory effects. MCPIP1 can be stimulated by many inflammatory factors such as LPS, IL-1β or MCP-1, and can be negatively regulated by down-regulating the expression of inflammatory cytokines such as IL-6 and IL-12p40. The mechanism of action of MCPIP1 is mainly to regulate the degradation of certain inflammatory cytokines or pre-miRNAs as RNases. In addition, MCPIP1 also acts as a deubiquitinating enzyme that targets TNF receptor-related proteins (TRAFs) and negatively regulates JNK and NF-κB signaling. The discovery of MCPIP1 molecules, gene and protein structure, biological functions, expression regulation and clinical application prospects are described in several aspects.
