目的:观察异丙酚对卡巴胆碱(CCh)引起的肠黏膜分泌的影响。方法利用 Ussing chamber 技术监测黏膜短路电流(Isc )的变化。结果CCh 引起大鼠回肠黏膜 Isc升高;异丙酚明显抑制 CCh 引起的大鼠回肠黏膜 Isc的变化,且呈现明显的浓度依赖效应(10-2~10-6 mol/L),这一抑制效应仅发生在浆膜层。钠离子通道阻断剂河豚毒素及 N 型胆碱能受体阻断剂六甲溴铵都不能影响异丙酚引起的抑制作用。结论静脉麻醉药异丙酚可抑制 CCh引起的肠黏膜分泌增加,其作用是通过抑制肠黏膜上皮基底侧的 M 型胆碱能受体信号途径实现。“,”Objective To explore the effect of propofol on carbachol(CCh)-induced chloride secretion in rat ileum mu-cosa.Methods Cch-induced chloride secretion was measured by Ussing chamber technique.Results Propofol atten-uated the CCh-stimulated increase in short-circuit current (Isc )of rat ileum mucosa in a dose-dependent manner (10 -2-10 -6 mol/L).The propofol-evoked inhibitory effect was serosal side-dependent.Furthermore,both TTX,a Na + channel blocker,and hexamethonium bromide,a nicotinic acetylcholine receptor antagonist,had no effect on propofol-evoked in-hibitory effect.Conclusion Propofol inhibites CCh-induced intestinal secretion via basolateral mAChR pathway.
