【目的】探讨在气道上皮鳞状化生过程中β-连接蛋白(β-cat)和维甲酸受体(RARβ)的表达和作用。【方法】选用初断乳SD雄性大鼠,随机分为正常对照组、维生素A缺乏9周组(VAD9周组)、维生素A缺乏13周组(VAD13周组),采用维生素A缺乏法复制气道上皮鳞状化生大鼠模型,观察各组大鼠气道上皮的形态学变化,采用原位杂交技术检测大鼠气道上皮β-cat mRNA的表达,免疫组化技术检测β-cat、RARβ蛋白表达。【结果】VAD9周组与VAD13周组大鼠气道上皮出现增生和鳞状化生,β-cat蛋白在胞质内的表达显著增加(P<0.001);β-cat mRNA的表达在各实验组和正常对照组间比较差异无显著性意义(P>0.05);RARβ在实验组的表达均较正常对照组显著减低(P<0.01)。【结论】在维生素A缺乏所致大鼠气道上皮鳞状化生过程中RARβ和β-cat表达异常,β-cat蛋白表达变化并不是其mRNA水平改变所致,可能因RARβ的表达减少,从而减弱其对β-cat/T细胞因子(TCF)信号的抑制,促进上皮鳞状化生而形成。

The expression of RARβ in the experimental group was significantly lower than that of the normal control group (P <0.01). 【Conclusion】 The expression of RARβ and β-cat in rat airway epithelial squamous metaplasia caused by vitamin A deficiency is abnormal. The change of β-cat protein expression is not caused by the change of mRNA level, which may be due to the decrease of RARβ expression, Thereby weakening its inhibition of β-cat / T cytokine (TCF) signaling and promoting the formation of epithelial squamous metaplasia.
