目的观察大鼠缺血区皮层Smac/Diablo的动态变化,并探讨其在缺血后神经元凋亡过程中的可能作用。方法采用RT-PCR,Western blot方法分别测定大鼠脑缺血后不同时间点在缺血区皮层Smac/Diablo mRNA和蛋白水平的表达水平。结果Smac/Diablo基因转录水平和蛋白表达的改变呈时间依赖性;RT-PCR显示模型组大鼠在脑缺血3h就有明显的Smac/Diablo mRNA表达上调,6h达高峰,持续至24h后逐渐下降,48h时恢复至缺血前水平;western blot显示在脑缺血后3h有Smac/Diablo蛋白表达的增加,6h时Smac/Diablo蛋白表达至高峰(P<0.01),48h基本恢复到缺血前水平。结论Smac/Diablo在脑缺血损伤后的病理生理过程中具有重要作用。
Objective To observe the dynamic changes of Smac / Diablo in ischemic cortex of rats and to explore its possible role in the process of neuronal apoptosis after ischemia. Methods The expression of Smac / Diablo mRNA and protein in ischemic cortex were detected by RT-PCR and Western blot respectively at different time points after cerebral ischemia. Results The transcriptional level of Smac / Diablo gene and the expression of Smac / Diablo gene were time-dependent. RT-PCR showed that the expression of Smac / Diablo mRNA was up-regulated in the model group 3h after cerebral ischemia and peaked at 6h, (P <0.01). The expression of Smac / Diablo protein peaked at 6h (P <0.01), and returned to the level of ischemia at 48h Before the level. Conclusion Smac / Diablo plays an important role in the pathophysiological process after cerebral ischemia.
