目的:探讨三黄泻心汤对重型颅脑外伤大鼠损伤区脑皮质核因子(NF-κB)、白细胞介素-6(IL-6)表达的影响。方法:Wistar大鼠70只(雄雌各半),随机分为正常对照组10只,模型组30只,中药组30只,后2组再按应激后6、24、72h共3个时相点平均分为3个亚组(即每组10只),除正常对照组外,其余各组予自由落体撞击法建立大鼠重型颅脑损伤模型,中药组予三黄泻心汤水煎液1mL/100g灌胃,其余组等体积生理盐水灌胃,每组分别在造模后6、24、72h 3个时相点取材,行光镜检查,并用免疫组化法检测不同时相点脑组织NF-κB及IL-6的表达。结果:IL-6、NF-κB着色于胞浆,在重型颅脑损伤大鼠各组脑组织中均有表达,24h阳性细胞数达到高峰,72小时开始缓慢下降。与正常对照组比较,模型各组,中药6、24h组大鼠损伤区皮质IL-6、NF-κB的表达含量较正常对照组明显增加(P<0.05,P<0.01);与模型72h组比较,中药72h组大鼠损伤区皮质IL-6、NF-κB的表达含量显著降低(P<0.05)。结论:三黄泻心汤抑制重型颅脑损伤大鼠脑组织核转录因子NF-κB、炎症因子IL-6的表达进而抑制炎症反应可能是其脑保护作用的机制之一。
Objective: To investigate the effect of San Huang Xie Xin Tang on the expression of nuclear factor (NF-κB) and interleukin-6 (IL-6) in cerebral cortex of rats with severe traumatic brain injury. Methods: Seventy male and female Wistar rats were randomly divided into normal control group (n = 10), model group (n = 30) and traditional Chinese medicine group (n = 30) The average points were divided into three subgroups (that is, each group of 10), in addition to the normal control group, the rest of the free fall into the body to establish a rat model of severe traumatic brain injury, Chinese medicine group Sanhuang Xiexin Decoction 1mL / 100g gavage, and the remaining group of equal volume of normal saline gavage, each group were made at 6,24,72 h after 3 hours of preparation, light microscopy and immunohistochemical detection of different time point brain Tissue NF-κB and IL-6 expression. Results: The staining of IL-6 and NF-κB in the cytoplasm was observed in all groups of rats with severe craniocerebral injury. The number of positive cells reached a peak at 24 hours and then decreased slowly at 72 hours. Compared with the normal control group, the expressions of IL-6 and NF-κB in the cortex of rats in the model group and the 6 and 24 h groups were significantly increased compared with the normal control group (P <0.05, P <0.01) Compared with the control group, the expression of IL-6 and NF-κB in the cortex of rats in the 72h group decreased significantly (P <0.05). Conclusion: Inhibition of inflammatory reaction by Sanhuang Xiexin Decoction in the brain of rats with severe craniocerebral injury may be one of the mechanisms of its protective effects on brain injury by inhibiting the expression of NF-κB and IL-6.
