目的通过建立类似于人脑胶质瘤的动物模型,观察颅内肿瘤生长特点。方法借助动物立体定向仪,将体外培养大鼠C6胶质瘤细胞(细胞数为3×105个,悬浮于无血清DMEM培养液60μL中)接种于Wistar大鼠左侧尾状核区。接种后观察实验大鼠的生存状态及肿瘤的生长特性,分别于接种后5、14 d进行MRI检查。在实验3、6、9、121、5 d时解剖标本,做组织病理学和GFAP免疫组化检查。结果该方法建立的胶质瘤动物模型在组织病理学上接近人脑胶质瘤.而且颅内生长稳定,成瘤率高,未见颅外转移病灶,实验周期短,重复性好。结论C6大鼠脑胶质瘤动物模型肿瘤生长及病理特征与人脑胶质瘤相似,可作为临床胶质瘤基础研究的理想模型。
Objective To establish an animal model similar to human glioma to observe the characteristics of intracranial tumor growth. Methods Rat C6 glioma cells (3 × 105 cells, suspended in serum-free DMEM medium 60 μL) were inoculated into left caudate nucleus of Wistar rats by using animal stereotactic instrument. After inoculation, the survival state of the rats and the growth characteristics of the tumor were observed. MRI examination was performed on the 5th and 14th day after inoculation. The specimens were dissected at 3, 6, 9, 121 and 5 d, and histopathology and GFAP immunohistochemistry were performed. Results The animal model of glioma established by this method was histopathologically close to human glioma, and had stable intracranial growth, high tumor formation rate, no extracranial metastatic lesions, short experimental period and good repeatability. Conclusion The C6 glioma animal model of tumor growth and pathological features similar to human glioma can be used as an ideal model for clinical glioma basic research.
