目的 :研究国产银杏内酯对脑缺血组织兴奋性氨基酸 (EAA)、抑制性氨基酸 (IAA)及脑含水量的影响。方法 :建立正常脑温大鼠大脑中动脉缺血 再灌注模型 ,用高效液相色谱荧光法检测脑组织谷氨酸 (Glu)、天门冬氨酸 (Asp)、甘氨酸 (Gly)、γ 氨基丁酸 (GABA)含量 ;用干湿质量法测定脑含水量。结果 :与对照组比较 :随银杏内酯剂量增加 ,Glu、Asp、Gly明显降低 ,GABA明显增高 ;治疗组脑含水量明显低于对照组。结论 :国产银杏内酯能抑制脑缺血后EAA过多释放 ,增强IAA释放 ,减轻脑水肿 ,有利于修复脑缺血引起的氨基酸失调。
Objective : To study the effects of domestic ginkgolide on excitatory amino acid (EAA), inhibitory amino acid (IAA) and brain water content in cerebral ischemic tissue. METHODS: A model of normal middle cerebral artery occlusion (I/R) was established in rats with cerebral ischemia. Glutamate (Glu), aspartate (Asp), glycine (Gly), and gamma aminobutyrin were detected by HPLC. Acid (GABA) content; Water content in brain was measured by dry-wet mass method. RESULTS: Compared with the control group, Glu, Asp, and Gly were significantly decreased, and GABA was significantly increased with increasing dose of ginkgolide; the brain water content of the treated group was significantly lower than that of the control group. Conclusion: Domestic ginkgolide can inhibit the release of EAA after cerebral ischemia, enhance the release of IAA, reduce cerebral edema, and is helpful to repair the amino acid imbalance caused by cerebral ischemia.
