目的检测红景天苷对小鼠宫颈癌移植瘤中Eag1表达情况的影响及其意义。方法建立小鼠宫颈癌U14细胞皮下移植瘤模型,将40只雌性昆明小鼠分为移植瘤组、顺铂组、红景天苷组和顺铂、红景天苷联合组,每天用游标卡尺测量肿块大小,采用电子天平称量瘤质量并计算抑瘤率,采用免疫组织化学法证实移植瘤为未分化鳞癌,取移植瘤组的瘤旁组织为对照组。采用RT-PCR和Western blot检测各组小鼠宫颈癌移植瘤中Eag1mRNA和蛋白的表达水平。结果皮下注射宫颈癌U14细胞建立昆明小鼠皮下移植瘤模型成瘤率100%。顺铂组、红景天苷组及联合组移植瘤的体积及瘤质量均低于移植瘤组,联合组移植瘤的体积及瘤质量明显低于顺铂组和红景天苷组(P<0.05);移植瘤组、顺铂组、联合组Eag1mRNA和蛋白表达明显高于对照组,顺铂组、红景天苷组、联合组Eag1mRNA和蛋白表达均低于移植瘤组,联合组Eag1表达水平低于顺铂组和红景天苷组,差异具有统计学意义(P<0.05)。结论红景天苷能抑制小鼠宫颈癌移植瘤的生长及Eag1表达,与顺铂联用效果更为显著。
Objective To detect the effect of salidroside on Eag1 expression in mouse cervical cancer xenografts and its significance. Methods A subcutaneous xenograft tumor model was established in U14 cells of mice with cervical cancer. Forty female Kunming mice were divided into transplanted tumor group, cisplatin group, salidroside group and cisplatin and salidroside combined group. The tumor mass was measured by electronic balance and the tumor inhibition rate was calculated. Immunohistochemistry was used to confirm that the xenograft tumor was undifferentiated squamous cell carcinoma, and the tumor-adjacent tissues in the transplanted tumor group were taken as the control group. The expression of Eag1 mRNA and protein in cervical cancer xenografts in each group was detected by RT-PCR and Western blot. Results The subcutaneous injection of cervical cancer U14 cells Kunming mouse subcutaneous tumor model of tumor formation rate of 100%. The volume and tumor mass of transplanted tumor in cisplatin group, salidroside group and combined group were lower than those in transplanted tumor group. The volume and tumor mass of transplanted tumor in combined group were significantly lower than those in cisplatin group and salidroside group (P < 0.05). The expression of Eag1 mRNA and protein in the xenografts, cisplatin and combination groups was significantly higher than that in the control group. The expressions of Eag1 mRNA and protein in the cisplatin group, salidroside group and the combination group were lower than those in the xenograft group and the combination group The level was lower than that of cisplatin group and salidroside group, the difference was statistically significant (P <0.05). Conclusions Salidroside can inhibit the growth of cervical cancer xenografts in mice and the expression of Eag1, which is more significant with cisplatin.
