目的 探讨内毒素脂多糖 (LPS)致新生大鼠肺出血时细胞间黏附分子 1(ICAM 1)和核因子κB抑制蛋白 (IκBα)表达的变化及其在肺出血发生机制中的作用。 方法 将 6 8只 7~ 10日龄Wistar大鼠分为LPS组和生理盐水组 (对照组 )。LPS组LPS 5mg/kg腹腔注射 ,按LPS注射后观察的时间分为 30min、1、2、4、8、16和 2 4h组 ;腹腔注射等量生理盐水作为对照组。对新生大鼠肺脏进行大体、光镜和肺超微结构的观察 ,分别采用Westernblot杂交和原位杂交法动态观察肺组织I CAM 1蛋白水平、mRNA表达及IκBα 表达的变化。 结果 注射LPS后 1h可引起新生大鼠明显的肺出血及炎性改变 ;肺组织ICAM 1蛋白表达逐渐增强 ,2h时明显高于对照组 (其吸光度A值分别为 178.13± 8.18、16 1.5 7± 12 .5 2 ,P <0 .0 5 ) ,至 2 4h时仍明显高于对照组 (吸光度A值为 192 .71± 16 .39,P <0 .0 1) ;ICAM 1mRNA的表达亦明显增强 ,其灰度值于 2h时明显降低 (分别为 10 7.17± 6 .2 3、111.4 5± 6 .14 ,P <0 .0 5 ) ,4h时与对照组比较差异有非常显著性 (其灰度值为 10 3.4 9±6 .4 5 ,P <0 .0 1) ;而于注射LPS后IκBα 表达很快减弱 ,1h时明显低于对照组 (其吸光度A值分别为171.5 3± 7.6 0和 182 .6 2± 6 .80 ,P <0 .0 5 ) ,8h时表达最
Objective To investigate the changes of intercellular adhesion molecule 1 (ICAM 1) and nuclear factor κB inhibitor (IκBα) expression in pulmonary hemorrhage induced by lipopolysaccharide (LPS) in neonatal rats and their roles in the pathogenesis of pulmonary hemorrhage. Methods Sixty-eight Wistar rats aged 7-10 days were divided into LPS group and normal saline group (control group). The rats in LPS group were injected intraperitoneally with LPS 5 mg / kg, and were divided into 30 min, 1, 2, 4, 8, 16 and 24 h groups according to the time of LPS injection. The rats were injected intraperitoneally with normal saline as the control group. The ultrastructure of the lungs of neonatal rats were observed, light microscopy and lung ultrastructure were observed. Western blot and in situ hybridization were used to observe the changes of I CAM 1 protein expression, mRNA expression and IκBα expression in lung tissues. Results One hour after injection of LPS, pulmonary hemorrhage and inflammatory changes were induced in neonatal rats. The expression of ICAM 1 protein in lung tissue was gradually increased at 2 hours and significantly higher than that in the control group (the absorbance A values were 178.13 ± 8.18 and 16 1.57 ± 12.52, P <0.05), still significantly higher than the control group at 24 hours (the absorbance A value was 192.71 ± 16.39, P <0.01), and the expression of ICAM 1 mRNA was also significant (P <0.05), and the gray value decreased significantly at 2h (10 7.17 ± 6.23,111.4 5 ± 6.14, P <0.05).
