目的探讨血必净注射液对大鼠下肢缺血再灌注损伤的保护作用,并进一步探索其最佳给药时间。方法制备大鼠下肢缺血再灌注模型(缺血2h,再灌注4h),将健康Wistar大鼠50只随机分为5组:A组为假手术组(n=10);B组为缺血前给药组(n=10);C组为缺血前给药对照组(n=10);D组为再灌注前给药组(n=10);E组为再灌注前给药对照组(n=10)。观察不同组间血清中超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量的变化。结果与假手术组比较,两对照组血清中SOD活性明显降低(P<0.01),MDA含量明显升高(P<0.01)。与对照组比较,给药组血清中SOD活性明显升高(P<0.01),MDA含量明显降低(P<0.01)。与D组比较,B组血清中SOD活性升高(P<0.05),MDA含量降低(P<0.05)。结论血必净可提高血清中SOD活性,降低MDA含量,对大鼠下肢缺血再灌注损伤有保护作用,在缺血前给予血必净效果更佳。
Objective To investigate the protective effect of Xuebijing injection on ischemia-reperfusion injury of lower extremities in rats and to explore its optimal administration time. Methods Fifty healthy Wistar rats were randomly divided into 5 groups: sham-operated group (n = 10), ischemia-reperfusion group B (N = 10); Group C was pre-ischemic control group (n = 10); Group D was reperfusion pre-administration group (n = 10); Group E was pre-reperfusion administration control Group (n = 10). The changes of serum superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in different groups were observed. Results Compared with the sham operation group, the SOD activity in the two control groups was significantly decreased (P <0.01) and MDA content was significantly increased (P <0.01). Compared with the control group, the activity of SOD in the serum of the treated group was significantly increased (P <0.01) and the content of MDA was significantly decreased (P <0.01). Compared with group D, serum SOD activity increased (P <0.05) and MDA content decreased (P <0.05). Conclusion Xuebijing can increase SOD activity in serum and decrease the content of MDA, which has a protective effect on ischemia-reperfusion injury of lower extremity in rats. It is more effective to give Xuebijing before ischemia.
