目的:制备聚乙二醇(PEG)化多西他赛白蛋白纳米粒(PEG-DANPs),比较市售多西他赛(艾素~,Aisu~)、多西他赛白蛋白纳米粒(DANPs)和PEG-DANPs对人非小细胞肺癌(NSCLC)体内外抗癌作用的差异,从而为临床治疗NSCLC提供一种新思路。方法:利用乳化挥发交联法制备DANPs,再将其进行PEG化制备出PEG-DANPs。将Aisu~,DANPs和PEG-DANPs分别进行体外溶血、体外释放实验,MTT法检测药物抑制NSCLC A549细胞增殖的情况,流式实验检测药物诱导癌细胞凋亡情况,激光共聚焦技术观察癌细胞对2种纳米粒制剂的摄取情况,最终比较三者治疗NSCLC的差异。结果:相比于Aisu~和DANPs,PEGDANPs具有明显的缓释效应,对机体红细胞破坏较少;PEG-DANPs能够更好地进入A549细胞,可以有效抑制A549细胞的增殖并诱导其凋亡。结论:PEG-DANPs是一种抗肿瘤细胞效果明显、不良反应较小的制剂,具有广阔的应用前景。
OBJECTIVE: To prepare polyethylene glycol (PEG) -mediated docetaxel albumin nanoparticles (PEG-DANPs) and to compare the efficacy and toxicity of docetaxel (Atsufloxacin, Aisu ~ ), docetaxel albumin nanoparticles (DANPs) and PEG-DNA on human non-small cell lung cancer (NSCLC) in vitro and in vivo anticancer effects, and thus provide a new idea for the clinical treatment of NSCLC. Methods: PEG-DANPs were prepared by emulsifying volatile cross-linking method and then pegylated. Aisu ~ , DANPs and PEG-DANPs were respectively in vitro hemolysis and in vitro release assay. MTT assay was used to detect the proliferation of NSCLC A549 cells. Flow cytometry was used to detect the apoptosis of drug-induced cancer cells. Laser confocal microscopy On the two kinds of nanoparticle preparations uptake, the final comparison of the three treatment of NSCLC differences. Results: Compared with Aisu ~ and DANPs, PEGDANPs had a sustained release effect and less damage to the body’s erythrocytes. PEG-DANPs could better enter A549 cells and effectively inhibit A549 cell proliferation and induce apoptosis. Conclusion: PEG-DANPs is an anti-tumor cell with obvious effect and small adverse reaction. It has broad application prospects.
