目的探讨乌司他丁(ulinastatin,UTI)对脓毒症大鼠心肌损伤的保护作用。方法63只SD大鼠随机分为假手术组(A组)、脓毒症组(B组)和乌司他丁组(C组),检测脓毒症前及脓毒症后3、6、12和24h大鼠血浆TNI、BNP水平和心肌MDA含量、SOD活性,以及UTI的影响,并观察心肌组织超微结构变化。结果脓毒症后3、6、12和24h大鼠血浆TNI、BNP水平以及心肌MDA含量均显著高于假手术组(P<0.05),心肌SOD活性低于假手术组(P<0.05);UTI组较脓毒症组血浆TNI、BNP、心肌MDA明显降低(P<0.05),心肌SOD活性明显升高(P<0.05);UTI组心肌组织超微结构的损害较脓毒症组明显减轻。结论脓毒症大鼠早期即发生明显的心肌损伤,乌司他丁能够显著减轻脓毒症时心肌损伤的程度,对脓毒症大鼠的心肌损伤有显著的保护作用。
Objective To investigate the protective effect of ulinastatin (UTI) on myocardial injury in septic rats. Methods Sixty-three SD rats were randomly divided into sham-operation group (A group), sepsis group (B group) and ulinastatin group (C group) The levels of plasma TNI and BNP, the content of MDA in myocardium, the activity of SOD and the activity of UTI in 12th and 24th hour rats were observed. The changes of myocardial ultrastructure were also observed. Results At 3, 6, 12 and 24 h after sepsis, the levels of plasma TNI, BNP and the content of MDA in myocardium were significantly higher than those in sham operation group and the sham operation group (P <0.05). In the UTI group, the levels of plasma TNI, BNP, MDA in myocardium were significantly lower than those in sepsis group (P <0.05), while the activity of SOD in myocardium was significantly increased (P <0.05). The ultrastructure damage in myocardium of UTI group was significantly lower than that in sepsis group . Conclusion Wistar rats have obvious myocardial injury in the early stage, and ulinastatin can significantly reduce the degree of myocardial injury in sepsis and have a significant protective effect on myocardial injury in septic rats.
