目的观察hNIS在不同病理类型的甲状腺组织中的表达情况,进而探讨hNIS表达水平与甲状腺疾病发生、发展的关系。方法收集临床甲状腺术后患者的组织块,包括桥本甲状腺炎3例,甲状腺瘤4例、结节性甲状腺肿4例、Graves病3例、亚急性甲状腺炎2例,分别采应用RT-PCR和Western bolt的方法检测不同病理类型的甲状腺组织中hNISmRNA及蛋白质的表达水平。结果桥本甲状腺炎hNIS表达明显降低,Graves hNIS表达明显增加,亚急性甲状腺炎、结节性甲状腺肿中的hNIS表达低于正常组织,腺瘤中的hNIS表达情况高于正常甲状腺组织。结论不同病理甲状腺组织hNIS在甲状腺疾病中发挥重要的作用,hNIS表达的改变,对甲状腺疾病的发生、发展有着十分重要的意义。
Objective To observe the expression of hNIS in different pathological types of thyroid tissue and to explore the relationship between the expression of hNIS and the occurrence and development of thyroid diseases. Methods Tissue blocks of patients with clinical thyroid surgery were collected, including 3 Hashimoto’s thyroiditis, 4 thyroid adenoma, 4 nodular goiter, 3 Graves disease and 2 subacute thyroiditis. RT-PCR Western blot was used to detect the expression of hNIS mRNA and protein in different pathological types of thyroid tissue. Results Hashimoto’s thyroiditis hNIS expression was significantly reduced, Graves hNIS expression was significantly increased, subacute thyroiditis, nodular goiter hNIS expression was lower than normal tissue, adenoma hNIS expression was higher than normal thyroid tissue. Conclusion hNIS plays an important role in thyroid diseases in different pathological thyroid tissues. The change of hNIS expression is of great significance for the occurrence and development of thyroid diseases.
