粗品肝素钠用FPA98C1树脂吸附,研究吸附液pH、盐浓度、温度对肝素钠效价和单位数的影响。结果表明,使用FPA98Cl树脂的最佳吸附工艺条件为:反应釜搅拌速度100 r/min,吸附温度56~57℃,吸附液pH7.9~8.0,盐浓度0.5 mol/L,m(树脂量)∶V(吸附液)=5 g/L,吸附时间8 h。在该工艺下,粗品肝素钠的平均效价达107.9 U/mg,平均单位数为68.83×103U,且其他理化结果良好。
The crude heparin sodium was adsorbed by FPA98C1 resin to study the effect of pH, salt concentration and temperature on heparin titer and unit number. The results showed that the optimal adsorption conditions of FPA98Cl resin were as follows: stirring speed of the reaction kettle 100 r / min, adsorption temperature 56 ~ 57 ℃, adsorption solution pH 7.9 ~ 8.0, salt concentration 0.5 mol / L, m (resin content) : V (adsorption solution) = 5 g / L, adsorption time 8 h. Under this process, the average titer of crude heparin sodium reached 107.9 U / mg, the average number of units was 68.83 × 103U, and the other physicochemical results were good.
