建立了液相色谱-串联质谱法测定大鼠尿液和粪便中的银杏内酯B。以白果内酯为内标,采用电喷雾电离源负离子模式(ESI-)、多反应监测(MRM)进行定量分析。监测离子对m/z 423.1→m/z 367.1(银杏内酯B)和m/z 325.2→m/z163.2(白果内酯)。银杏内酯B在1~250?g/ml(尿液)和1.6~400.0?g/g(粪便)范围内线性关系良好,方法回收率为81.8%~113.8%(尿液)和83.5%~115.2%(粪便),日内、日间RSD小于9.7%(尿液)和10.4%(粪便)。采用该法考察大鼠静脉注射银杏内酯B后的排泄动力学,48 h尿液与粪便合计累积排泄率为54.09%。
A liquid chromatography-tandem mass spectrometry was developed for the determination of ginkgolide B in rat urine and feces. Using bilobalide as internal standard, electrospray ionization negative ion mode (ESI-) and multiple reaction monitoring (MRM) were used for quantitative analysis. Ion pair pairs m / z 423.1 → m / z 367.1 (ginkgolide B) and m / z 325.2 → m / z 163.2 (bilobalide) were monitored. Ginkgolide B had a good linearity in the range of 1 ~ 250 μg / ml (urine) and 1.6 ~ 400.0 μg / g (stool), and the recoveries were 81.8% -1 113.8% (urine) and 83.5% 115.2% (droppings), intraday RSD less than 9.7% (urine) and 10.4% (droppings) during the day. The method was used to investigate the excretion kinetics of ginkgolide B after intravenous injection of Ginkgolide B in rats. The combined cumulative excretion rate of urine and faeces at 48 h was 54.09%.
