目的探讨子宫内膜异位症与健康对照组之间的血清差异表达蛋白,为子宫内膜异位症早期诊断提供生物标志物的检测方法。方法选择临床明确诊断的子宫内膜异位症患者以及健康妇女(对照组)各20例,采集其血清利用同位素标记相对和绝对定量(iTRAQ)技术检测其血清差异蛋白,并进行功能注释及生物信息分析。并从中选择明显相关的蛋白,利用Western-blot进行验证。结果与对照组相比,子宫内膜异位症患者血清中有29个蛋白异常表达,其中4个蛋白表达明显降低,25个蛋白表达明显升高。Western-blot验证结果与iTRAQ结果相符。结论证实了iTRAQ技术对于筛选生物标记物具有实用性。CD44,纤维蛋白原-α和纤连蛋白1等参与细胞粘附的分子在子宫内膜异位症的发病机制中起作用。
Objective To investigate the differential expression of serum proteins between endometriosis and healthy controls and to provide biomarkers for the early diagnosis of endometriosis. Methods Twenty patients with clinically diagnosed endometriosis and 20 healthy women (control group) were selected. Serum were collected for serum differential protein detection using isotope labeled relative and absolute quantification (iTRAQ), and functional annotation and bioinformatics Information Analysis. And select the obvious related proteins, using Western-blot to verify. Results Compared with the control group, 29 patients with endometriosis serum abnormal expression, of which 4 protein was significantly reduced, 25 protein was significantly increased. Western-blot results are in good agreement with iTRAQ results. The conclusion confirms that the iTRAQ technique is practical for screening biomarkers. Molecules involved in cell adhesion such as CD44, fibrinogen-α and fibronectin 1 play a role in the pathogenesis of endometriosis.
