目的 探讨端粒酶蛋白催化亚单位 (hTERT)基因mRNA及其蛋白在胃癌发生、发展中的重要作用以及在胃癌诊断中的意义。方法 应用原位杂交方法检测 5 3例胃癌标本中hTERTmRNA的表达 ,应用免疫组化S P法检测相同标本中hTERT蛋白的表达。结果 5 3例胃癌标本中 ,hTERTmRNA阳性 41例 (77% ) ,其中低分化腺癌阳性 18例 (82 % ) ,高分化腺癌阳性 2 1例 (81% ) ,黏液腺癌及黏液细胞癌阳性 2例 (2 / 5 ) ;hTERT蛋白阳性共 46例 (87% ) ,其中低分化腺癌阳性 2 0例 (91% ) ,高分化腺癌阳性 2 5例(96 % ) ,黏液腺癌及黏液细胞癌阳性 1例 (1/ 5 ) ;hTERTmR NA及hTERT蛋白的表达在低分化腺癌组与高分化腺癌组之间差异未见显著性 (P >0 0 5 ) ,但hTERTmRNA与hTERT蛋白之间呈正相关 (rS=0 6 2 3,P <0 0 1)。结论 hTERTmRNA及蛋白表达的检测有可能为胃癌的诊断及鉴别诊断提供一种新的形态学的指标
Objective To investigate the role of telomerase catalytic subunit (hTERT) gene mRNA and its protein in the occurrence and development of gastric cancer and its significance in the diagnosis of gastric cancer. Methods The expression of hTERT mRNA in 53 gastric cancer samples was detected by in situ hybridization. The expression of hTERT protein in the same samples was detected by immunohistochemical SP method. Results The positive hTERT mRNA was found in 41 cases (77%) of 53 gastric cancer samples, of which 18 (82%) were positive for poorly differentiated adenocarcinoma and 21 (81%) for well-differentiated adenocarcinoma. Muco-adenocarcinoma and mucinous cell carcinoma Positive in 2 cases (2/5); hTERT protein positive in 46 cases (87%), including poorly differentiated adenocarcinoma 20 cases (91%), well-differentiated adenocarcinoma positive 25 cases (96%), (1/5). The expression of hTERTmR NA and hTERT protein had no significant difference between poorly differentiated adenocarcinoma and well-differentiated adenocarcinoma (P> 0.05), but hTERTmRNA and hTERT protein was positively correlated (rS = 0 6 2 3, P <0 0 1). Conclusion The detection of hTERT mRNA and protein expression may provide a new morphological index for the diagnosis and differential diagnosis of gastric cancer
