防止组织培养中污染的技术措施

组织培养液具备适合于哺乳类细胞生长的各种营养成份。主要有无机盐,氨基酸和维生素组成。多数还须加入血清才能生长,血清的机制尚不十分明了。不含血清的化学综合培养基可适合于某些异倍体细胞的生长。许多学者曾创立了一些适于某特定细胞生长的培养液,所有培养液共同特点是给微生物提供了极好的生长条件。 在组织培养操作过程中,如接种或再培养均需打开容器,培养的温度又适宜,极易造成污染。何况离体细胞不如在机体中具有物理或化学的防御机能。例如人类皮肤等防止感染,就具有下列特点:①干燥表皮细胞;②多层化;③表层不断脱落;④pH在4.0—6.0,平均4.85;⑤表面披以油质,防止浸湿与浸溶;⑥角质化防止擦伤;⑦披以表 Tissue culture fluid has a variety of nutrients suitable for mammalian cell growth.
