目的:探讨杂色曲霉素(ST)对体外培养的人外周血单个核细胞(HPBMc)抗原加工递呈相关基因TAP1、LMP2表达的影响。方法:采用RT-PCR和FCM方法,分析5种浓度ST处理对体外培养的HPBMcTAP1、LMP2基因在mRNA和蛋白水平表达的影响。结果:RT-PCR检测结果表明,ST处理24h后,低浓度ST组(0.125mg/L、0.25mg/L)HPBMcTAP1mRNA相对表达量明显高于对照组,而较高浓度组(0.5mg/L、1mg/L和2mg/L)TAP1mRNA则明显低于对照组,尤以1mg/L和2mg/L组为著。FCM定量分析表明,低浓度ST组HPBMcTAP1蛋白平均表达量略低于对照组,但无统计学意义,而较高浓度组TAP1表达则明显低于对照组(P<0.05)。在0.125mg/L到1mg/L浓度范围内,各ST处理组HPBMcLMP2蛋白表达量和mRNA的相对表达量均高于对照组。结论:ST对体外培养的HPBMcTAP1mRNA表达和蛋白表达的影响按ST剂量不同而变化,在0.125~1mg/L浓度范围内ST在mRNA及蛋白水平均可剂量依赖地提高体外培养的HPBMcLMP2的表达。
Objective: To investigate the effect of streptozotocin (ST) on the expression of TAP1 and LMP2 in human peripheral blood mononuclear cells (HPBMc) antigen in vitro. Methods: The mRNA and protein levels of HPBMcTAP1 and LMP2 genes cultured in vitro were analyzed by RT-PCR and FCM. Results: RT-PCR results showed that the relative expression level of HPBMcTAP1 mRNA in ST group (0.125mg / L, 0.25mg / L) was significantly higher than that in control group 1mg / L and 2mg / L) TAP1mRNA was significantly lower than the control group, especially 1mg / L and 2mg / L group. FCM quantitative analysis showed that the average expression level of HPBMcTAP1 protein in ST group was slightly lower than that in control group, but there was no statistical significance, while the TAP1 expression in higher concentration group was significantly lower than that in control group (P <0.05). In the concentration range of 0.125 mg / L to 1 mg / L, the expression levels of HPBMcLMP2 protein and mRNA in each ST treatment group were higher than those in the control group. CONCLUSION: The effect of ST on the expression and protein expression of HPBMcTAP1 in vitro varies with the dose of ST. ST can increase the expression of HPBMcLMP2 in a dose-dependent manner at mRNA and protein levels in the range of 0.125-1mg / L.
