目的 通过观察雾化吸入三氧化二砷(As2O3)对哮喘豚鼠气道嗜酸粒细胞的病理改变,探讨As2O3剂雾化吸入的平喘治疗的可行性。方法 采用鸡卵蛋白(OVA)复制哮喘豚鼠模型,分组给予不同剂量As2O3喷射雾化吸入;同时设生理盐水雾化吸入组、大剂量的As2O3腹腔注射组对照,均用药7d后比较支气管粘膜下层的嗜酸粒细胞(EOS)的情况。结果 小剂量的As2O3(1.0和2.0mg/kg·d-1)雾化吸入组支气管粘膜下层的EOS明显减少,与哮喘组比较差异有高度显著性(P<0.01);与较大剂量的As2O3(5.0mg/kg·d-1)腹腔注射组比较差异无显著性(P>0.05)。结论 小剂量的As2O3雾化吸入能够显著减少哮喘豚鼠气道中的EOS,从而减轻哮喘的气道炎症,短期使用可以相对安全而有效地达到治疗哮喘的目的。
Objective To observe the pathological changes of airway eosinophils in asthmatic guinea pigs by inhalation of arsenic trioxide (As2O3) and explore the feasibility of antiasthmatic treatment with As2O3 inhalation. Methods Asthmatic guinea pig model was induced by ovalbumin (OVA), and inhaled by different doses of As2O3 sprayed in groups. At the same time, normal saline inhalation group and high dose of As2O3 intraperitoneal injection group were used for comparison. After 7 days, bronchial submucosa Eosinophils (EOS) situation. Results The EOS of bronchial submucosa of atomized inhalation group with low dose of As2O3 (1.0 and 2.0 mg / kg · d-1) was significantly lower than that of asthma group (P <0.01). Compared with the higher dose of As2O3 (5.0mg / kg · d-1) intraperitoneal injection group was no significant difference (P> 0.05). Conclusion As2O3 inhalation can significantly reduce the EOS in the airway of asthmatic guinea pigs and relieve the airway inflammation of asthma. Short-term use can safely and effectively treat asthma.
